Studies on the Clonal Multiplication of Carnation through the in Vitro Cuttings


Abstract
This study was performed to establish for clonal multiplication of spray carnation (cv. Scarlet Bell) through the in vitro cuttings. Node, leaf bud and shoot tip were separated from plantlet subcultured on solid medium of Murashige and Skoog (MS) without growth regulators for about one year in vitro. And then these materials were transplanted and subcultured on solid medium of MS with NAA and BA of various concentration. For clonal multiplication node, leaf bud and shoot tips were employable as subculture materials of carnation, and multiplication ability was compared favorably with axillary bud growth promotion by liquid culture. After a month transferred organ subcultured was made to multiple shoot; shoot number in node, leaf bud and shoot tip were 7.9, 6.0 and 7.3 in the most effective experimental plot respectively. That is, exogenous growth regulators were more required in leaf bud than in node or shoot tip. The most effective plots in shoot number are as follows; node, leaf bud and shoot tip were NAA 0.01ppm£«BA 1.0 ppm, NAA 1.0ppm£«BA 1.0ppm, and NAA 0.01ppm respectively.
